If you can read the BIOS on the chip, and a saved copy of it is an identical match for the one you flashed on it, then you can be 100% sure the connection with the chip and clip is good, and that a new flash will work as it should(unless the connection is disturbed).
You're doing all this first because a poor connection will cause errors during the flashing process. It makes things MUCH simpler as far as making sure 2 files are identical(it does the work for you). I suggest you familiarize yourself with how to use a hex editor(such as HxD), and use it to do a data comparison analysis(a built-in function for automatically comparing 2 files side-by-side) with the BIOS you flashed on the chip previously and a copy you can save after reading it with the programmer. If you don't see the exact same values as with another hex editor you have a poor connection. It should look exactly as it would in any hex editor.
